A Mechanism of Excessive Accumulation of Abomasal Gas in Vagotomized Cattle Determined using Fluoroscopy OAK
ITOH, Megumi; SASAKI, Naoki; KAWAMOTO, Satoshi; YAMADA, Haruo; INOKUMA, Hisashi; 佐々木, 直樹; 猪熊, 壽.
To better understand the mechanism of excessive gas accumulation in the abomasum in bovine abomasal displacement, we performed gastric fluoroscopy in vagotomized cattle. Fifteen 6-month-old Holstein steers were divided into three groups: a non-vagotomized control group (Group C; n=5), a ventral thoraco-vagotomized group (Group V; n=5), and a dorsal and ventral thoraco-vagotomized group (Group DV; n=5). These groups were examined by fluoroscopy before and during a 5-week observation period after surgery. In Group C, no change was observed throughout the observation period. In Group DV, immediately after surgery, reticuloruminal motility was completely absent and ruminal distention was seen. Two weeks after surgery, abnormal reticulum motility and increased...

SASAKI, Naoki; TSUZUKI, Nao; YAMADA, Michiaki; MINAMI, Takuto; YAMADA, Haruo; 佐々木, 直樹.
This study aimed to establish a standard dose and sample collection time for 13C phenylalanine and 13C-Dipeptide breath test in horses. To evaluate dose-dependent effects, healthy horses received 2.5 mg/kg, 5 mg/kg, and 10 mg/kg 13C phenylalanine dissolved in 1 ml/kg distilled water and 1.25 mg/kg, 2.5 mg/kg, and 5 mg/kg 13C dipeptide dissolved in 2 ml/kg distilled water. Tmax was observed during the sample collection time. For 13C phenylalanine, the standard deviation of Cmax at 5 mg/kg was lower than that of 10 mg/kg. For 13C dipeptide, the standard deviation of Tmax was the lowest at 5 mg/kg. This study revealed that an optimal dose for breath tests with 13C phenylalanine and 13C dipeptide may be 5 mg/kg in horses.

Basic Assessment of Diagnosis by MDCT in Horses OAK
SASAKI, Naoki; KIKUCHI, Takuya; YAMADA, Kazutaka; HIGUCHI, Tohru; SATO, Motoyoshi; YAMADA, Haruo; 佐々木, 直樹; 山田, 一孝.
In this equine study, we used an MDCT unit and a trestle for large animals to assess the utility of MDCT in the diagnosis locomotor system disorders and problems in the head and spine. We used a mature thoroughbred horse under general anesthesia and 5 specimens. For specimens, MDCT images were compared with other images taken by a computed radiography X-ray unit. In the live horse, use of the trestle for large animals enabled scanning under general anesthesia. In specimens, MDCT created 3D images of the fractured lesions and evaluation of multiple cross-sectional and sagittal images was possible. Further, multiple sagittal images and virtual endoscopic images revealed details of a spinal canal stenosis in the swayback case. These results indicate that MDCT...

Clinical Applications of Electrointestinography in the Horse OAK
SASAKI, Naoki; LEE, Inhyung; AYUKAWA, Yu; YAMADA, Haruo; 佐々木, 直樹.
It is thought that in order to effectively treat gastrointestinal dysfunction, it should be detected at an early stage. The present study aimed to observe the movement via Electrointestinography (EIG) in the small intestine, cecum, right ventral colon and right dorsal colon, and to examine the procedure for assessing gastrointestinal function. Seven healthy thoroughbred horses were used. They were allowed food twice a day. EIG was measured at ten-minute intervals over a 24 hr period, and running spectral analysis was used. As a result, it was clear that EIG could non-invasively assess gastrointestinal function by setting the cut-off value of the frequency band to 1.8-12.0 cpm for a part of the digestive tract which is in a comparatively fixed position. The...

Effects of Orally Administrated Mosapride, a 5-HT4 Receptor Agonist on the Digestive Tract of Horses OAK
SASAKI, Naoki; LEE, Inhyung; AYUKAWA, Yu; YAMADA, Haruo; 佐々木, 直樹.
The effects of mosapride, a 5-hydroxytryptamine 4 (5-HT4) receptor agonist, on the motility of horse small intestine were examined by means of electrointestinography (EIG). Six adult healthy thoroughbreds were used, in which EIG signals from the small intestine were measured by a Degetorappar EGG system. Mosapride, dissolved in 200 ml water, was orally administered at a dose of 2 mg/kg. The total power of EIG signals at a frequency band of 3 cpm was 419.1 ± 122.0 (μV) 2 × cpm (n=6) before, and 2,093.6 ± 850.0 (μV)2 × cpm (n=6) after the drug administration, the latter mean value being significantly greater than the former. This result suggests that mosapride has a prokinetic action on the motility of horse small intestine.

Evaluation of Techniques That Measure Hydrogen and Methane Levels in Breaths of Horses with Gastrointestinal Diseases OAK
SASAKI, Naoki; FUKUNAKA, Morito; YAMADA, Haruo; SENBA, Hiroyuki; HIGUCHI, Tohru; 佐々木, 直樹.
Techniques for measuring hydrogen (H2) and methane (CH4) levels in breaths were assessed in horses with gastrointestinal (GI) diseases. A total of 31 horses were studied: 11 healthy horses (Group A), 10 horses with GI diseases (Group B), and 10 horses with diseases without GI involvement (Group C). Expired gases were measured using gas chromatography. CH4 levels in Group A were significantly lower than those in Group B. In Group B, CH4 levels were significantly higher after treatment. Fluctuations in H2 and CH4 levels in breath were associated with changes in GI function.

Evaluation of the Cranial Cruciate Ligament Repair System® in Surgery for Laryngeal Hemiplegia in Heavy Draft Horses OAK
SASAKI, Naoki; MORITA, Yoshinori; MORIYAMA, Tomoe; YAMADA, Haruo; 佐々木, 直樹.
The purpose of this study was to evaluate the effect of the canine cranial cruciate ligament repair system on laryngeal hemiplegia in heavy draft horses. Twenty-four heavy draft horses diagnosed with grade 4 laryngeal hemiplegia were allocated to either the prosthetic laryngoplasty (PL) group (n=14) or a canine cranial cruciate ligament repair system (CCCLRS) group (n=10). Right to left angle quotients (RLQs) of abductions of the arytenoid cartilages were endoscopically evaluated before and after surgery. Post-operative RLQs in the CCCLRS group were significantly lower than those of the PL group (P<0.01). The canine cranial cruciate ligament repair system was revealed to be a good surgical procedure for laryngeal hemiplegia in heavy draft horses.

A 17-year-old, gray Thoroughbred gelding was presented with recurrent epistaxis following exercise for over one year. Multidetector-row computed tomographic (MDCT) examination of the head performed under general anesthesia indicated the presence of malignant melanoma. MDCT examination revealed the exact location and size of masses in the right guttural pouch first identified by endoscopy and in the left guttural pouch unidentified by endoscopy. MDCT examination revealed the presence of a blood clot that filled the right maxillary sinus. The horse was euthanized due to poor prognosis as a riding horse and the owner's decision, and MDCT findings were confirmed by postmortem examination. MDCT is suggested as a useful technique for diagnosis of head problems...

Medicinal Treatment to Equine Gastrointestinal Dysfunctions OAK
SASAKI, Naoki; YAMADA, Haruo; HARA, Shigeo; 佐々木, 直樹.
This study was designed to investigate gastrointestinal dysfunctions, especially the effects of gastrointestinal prokinetic agents on digestive disorders involving lowered motility. The effects of α2-adrenergic agonists for treating acute abdominal disorders involving intestinal torsion in horses were also investigated. The results are as follows: (1) It appeared that the functions of migrating contractions (MC) in horses were to transfer the contents of the digestive tract as soon as possible to the cecum and colon, in addition to maintaining homeostatis in the digestive tract. (2) Motilin was seen to be involved in the regulation mechanism of intestinal motility in the horse as well as in humans and dogs, and brought about MC. (3) The intravenous...

OKAMURA, Koichi; SASAKI, Naoki; FUKUNAKA, Morito; YAMADA, Haruo; INOKUMA, Hisashi; 佐々木, 直樹; 猪熊, 壽.
The prokinetic effect of the 5-HT4 receptor agonist mosapride was evaluated in seven healthy thoroughbreds. Mosapride was orally administerd at doses of 0.5, 1.0 or 1.5 mg/kg. The breath 13CO2/12CO2 rate (Δ13CO2), an indirect indicator for the rate of gastric empting, was measured at appropriate points for 4 hr after drug administration. There was a significant increase compared with the control value at 15, 20 and 165 min for 0.5 mg/kg, 30 min at 1.0 mg/kg and 165 min for 1.5 mg/kg. The results suggest that mosapride may facilitate the gastric emptying in horses.
